Non-ionic detergents and hypotonic treatment lyse erythrocytes and leukocytes membranes, facilitating the accumulation of leukocytes-nuclei in the pellet. This treatment removes most of the cellular proteins and DNA is quickly released (in 15-20 mins) and partially purified. The DNA is partially fragmented so is ideal for PCR applications, but not for Southern blot or Sequencing.
